ABSTRACT

Hemoplasmas are bacteria able to adhere themselves loosely to the plasma membrane of erythrocytes and may parasitize several species of mammals. There are three known species of hemoplasmas that parasitize domestic and wild cats: Mycoplasma haemofelis, 'Candidatus Mycoplasma haemominutum' and 'Candidatus Mycoplasma turicensis'. Dogs are infected by at least two species of hemoplasmas: 'Candidatus Mycoplasma haematoparvum' and Mycoplasma haemocanis. The hemoplasmoses are very important in veterinary clinics, either because of its worldwide distribution and severity of clinical signs, depending on parasite species and host immune competence, or due to its zoonotic potential and capability of infecting endangered species. This study set out to investigate which hemoplasmas species parasitize different captive wild carnivores in order to clarify the epidemiology of hemoplasmoses in wild animals. Furthermore, the research intended to characterize the hematological changes caused by different species of hemotropic mycoplasmas infection in order to establish their clinical importance to wild species and the capacity of these species to become a reservoir of studied agents. Samples of 33 wild felids and 18 wild canids were investigated using polymerase chain reaction (PCR) to detect hemoplasmas DNA and it was observed that the occurrence of infection in these species is 45.5% and 83.3%, respectively. Factors such as age, gender or anaemia are not more frequent in animals positive for the infection. Therefore, it is concluded that infection caused by hemoplasmas in wild carnivores has high prevalence, and either agent pathogenicity is low, or chronic stage is more frequent, resulting in a low rate of diagnosis.(AU)


Hemoplasmas são bactérias capazes de aderir frouxamente à membrana plasmática de eritrócitos e que podem parasitar diversas espécies de mamíferos. São conhecidas três espécies de hemoplasmas que parasitam felídeos domésticos e selvagens: Mycoplasma haemofelis, 'Candidatus Mycoplasma haemominutum' and 'Candidatus Mycoplasma turicensis'. Cães são infectados por ao menos duas espécies de hemoplasmas: Candidatus Mycoplasma haematoparvum' and Mycoplasma haemocanis. As hemoplasmoses são de grande importância na clínica veterinária, tanto pela sua distribuição ubíqua e severidade dos sinais clínicos, a depender da espécie do parasita e imunocompetência do hospedeiro, quanto pelo seu potencial zoonótico e capacidade de infectar espécies ameaçadas. Este estudo visa investigar quais espécies de hemoplasmas parasitam diferentes carnívoros selvagens de cativeiro, a fim de esclarecer a epidemiologia das hemoplasmoses em animais selvagens. Além disso, o trabalho objetivou caracterizar as alterações hematológicas causadas pela infecção por diferentes espécies de micoplasmas hemotrópicos visando estabelecer sua importância clínica para espécies selvagens e a capacidade destas espécies de se tornar reservatórios dos agentes estudados. Amostras de 33 felídeos selvagens e de 18 canídeos selvagens foram investigadas por meio da reação em cadeia da polimerase (RCP) para detectar o DNA dos agentes e foi observado que a ocorrência da infecção por hemoplasmas nestas espécies é de 45,5% e 83,3%, respectivamente. Fatores como idade, sexo ou anemia não são mais frequentes em animais positivos para a infecção. Dessa forma, conclui-se que a infecção causada por hemoplasmas em carnívoros selvagens possui alta prevalência, no entanto ou a patogenicidade dos agentes é baixa ou o estágio crônico da infecção é mais frequente, resultando em uma baixa frequência diagnóstica.(AU)

Objective: To survey hemoplasmas infection in free ranging non-human primates from 8 provinces in Thailand. Methods: DNA from ethylenediaminetetraacetic acid blood of 262 free ranging non-human primates were identified as hemoplasmas using PCR and phylogenetic analysis based on 16S rRNA and rnpB genes. Results: A total of 148 non-human primates (56.49%) were determined positive for Candidatus Mycoplasma haemomacaque, including 125 Macaca fascicularis and 23 Macaca mulatta. Hemoplasmas can cause anemia in monkey but all positive samples were healthy. The positive rates in male and female non-human primates were not significantly different. Conclusions: Candidatus Mycoplasma infection is prevalent in free ranging Macaca fascicularis and Macaca mulatta in Thailand.

ABSTRACT

In this study, we examine prevalences of three infectious pathogens with different transmission modes (Bartonella henselae, hemoplasma, and Toxoplasma gondii) in feral cats from urban and rural habitats. Infection status of the three pathogens in blood samples (n = 117) was determined through molecular or serological diagnostic methods. Overall prevalence of hemoplasma, Toxoplasma gondii, and Bartonella henselae was 47.9%, 50%, and 35.7%, respectively. Comparing the two habitats, only seroprevalence of Bartonella henselae was significantly higher in urban cats. Based on the results, we discuss how pathogens with distinct transmission modes may show different prevalence between urban and rural habitat types.

Abstract Hemotropic mycoplasmas in dogs, such as Mycoplasma haemocanis, have been described worldwide. Recently, these pathogens have been reported to be causative agent of zoonosis. It is known that its transmission may occur through the action of blood-sucking arthropods (e.g. ticks or fleas), through blood transfusion, contaminated fomites and/or transplacentally. In Brazil, M. haemocanis is present in practically all regions and the tick Rhipicephalus sanguineus sensu lato is suspected the main vector. In the municipality of Campo Grande, state of Mato Grosso do Sul, there is little information about infection of dogs by M. haemocanis, or on the main epidemiological features associated with it. Thus, the aim of the present study was to determine the occurrence of M. haemocanis among dogs infested by ticks and to assess possible associations with some epidemiological factors. The polymerase chain reaction (PCR) and DNA sequencing were used to analyze dog blood samples (n = 94). DNA from M. haemocanis was detected in four samples. No significant associations were observed with any epidemiological parameter analyzed here. However, the results from this study confirm that this pathogen is circulating in this region and should be considered in the differential diagnosis of diseases among anemic dogs.


Resumo Micoplasmas hemotrópicos de cães, como Mycoplasma haemocanis, já foram descritos em todo o mundo. Recentemente, esses patógenos têm sido apontados como causadores de zoonoses. É sabido que a transmissão pode ocorrer devido à ação de artrópodes sugadores de sangue (carrapatos, pulgas), transfusão sanguínea e/ou fômites contaminados e por via transplacentária. No Brasil, Mycoplasma haemocanis está presente em praticamente todas as regiões, e o carrapato Rhipicephalus sanguineus sensu lato é suspeito como principal vetor. No município de Campo Grande, Estado de Mato Grosso do Sul, Brasil não existem muitas informações acerca de infecções de cães por M. haemocanis, assim como quais são os principais aspectos epidemiológicos associados a este patógeno. Assim, o objetivo, no presente estudo, foi determinar a ocorrência de M. haemocanis em cães infestados por carrapatos e analisar possíveis associações com alguns fatores epidemiológicos. A Reação em Cadeia da Polimerase (PCR) e o sequenciamento de DNA foram utilizados para analisar amostras de sangue de cães (n = 94). DNA de M. haemocanis foi identificado em quatro amostras. Não foram observadas associações significativas com qualquer parâmetro epidemiológico analisado. No entanto, os resultados deste estudo confirmam que esse patógeno está circulando na região e deve ser considerado no diagnóstico diferencial de causas de anemia em cães.

SUMMARYThe aims of this study were to determine the prevalence of hemoplasmas in a rural Brazilian settlement's population of human beings, their dogs and horses, highly exposed to tick bites; to identify the tick species parasitizing dogs and horses, and analyze factors associated with their infection. Blood samples from 132 dogs, 16 horses and 100 humans were screened using a pan-hemoplasma SYBR green real-time PCR assay followed by a species-specific TaqMan real-time PCR. A total of 59/132 (44.7%) dog samples were positive for hemoplasmas (21 Mycoplasma haemocanisalone, 12 ' CandidatusMycoplasma haematoparvum' alone and 21 both). Only 1/100 (1.0%) human sample was positive by qPCR SYBR green, with no successful amplification of 16S rRNA or 23 rRNA genes despite multiple attempts. All horse samples were negative. Dogs >1 year of age were more likely to be positive for hemoplasmas ( p= 0.0014). In conclusion, although canine hemoplasma infection was highly prevalent, cross-species hemoplasma transmission was not observed, and therefore may not frequently occur despite overexposure of agents and vectors.


RESUMOOs objetivos deste estudo foram determinar a prevalência de hemoplasmas numa população restrita de cães, equinos e humanos altamente expostos a picadas de carrapatos em assentamento rural brasileiro; identificar as espécies de carrapatos parasitando cães e equinos, e analisar os fatores associados à infecção. Amostras de sangue de 132 cães, 16 cavalos e 100 humanos foram avaliadas utilizando um protocolo pan-hemoplasma em PCR quantitativas em tempo real (qPCR) com SYBR green, seguido de qPCR TaqMan espécie-específicos. Cinquenta e nove/132 (44,7%) cães foram positivos para hemoplasmas (21 Mycoplasma haemocanis, 12 ' Candidatus Mycoplasmahaematoparvum' e 21 para ambos). Uma amostra humana do total de 100 (1%) foi positiva pelo qPCR SYBR green, mas os genes 16S rRNA ou 23S rRNA não foram amplificados com sucesso, apesar de inúmeras tentativas. Todas as amostras de cavalos foram negativas. Cães > 1 ano apresentaram mais chance de serem positivos para hemoplasmas ( p= 0,0014). Concluindo, embora infecções por hemoplasmas caninos sejam altamente prevalentes, a transmissão de hemoplasmas entre espécies não foi observada, e desta forma podem não ocorrer de forma frequente apesar da alta exposição aos agentes e vetores.

ABSTRACT

This study examined the occurrence of Anaplasma spp. and hemoplasma infection in leopard cats, Prionailurus bengalensis euptilurus, in Korea. Twenty-nine biological samples were tested by molecular analysis. Two (6.9%) and eight (27.6%) tested specimens were positive for Anaplasma bovis and hemoplasma infection, respectively. Based on our results, Anaplasma/Ehrlichia spp. and hemoplasma are regularly infecting leopard cat populations of Korea. Considering their endangered status, regular monitoring of infection by arthropod-borne pathogens known to cause clinical symptoms in feline hosts such as Anaplasma/Ehrlichia spp. and hemoplasma would be crucial as part of ongoing conservation efforts.

ABSTRACT

Three hemoplasma species are recognized in domestic cats: Mycoplasma haemofelis, ‘Candidatus Mycoplasma haemominutum’ and ‘Candidatus Mycoplasma turicensis’. We report the prevalence and hematological abnormalities of hemoplasma infection in 369 domestic cats from three different populations (blood donors, hospitalized cats and shelter cats) from Southern Brazil. Complete blood counts were performed at the time of blood collection, and DNA was extracted and tested by conventional PCR for each hemoplasma species. A total of 79 samples (21.40%) were positive for at least one species. The most prevalent hemoplasma was ‘Candidatus Mycoplasma haemominutum’, with 50/369 (13.55%) positive cats, followed by ‘Candidatus Mycoplasma turicensis’, 10/369 (2.71%), and Mycoplasma haemofelis, 8/369 (2.16%). Mycoplasma haemofelis and ‘Candidatus Mycoplasma haemominutum’ coinfection was observed in 4/369 (1.08%), whereas ‘Candidatus Mycoplasma haemominutum’ and ‘Candidatus Mycoplasma turicensis’ in 5/369 (1.35%). Three cats (0.81%) were infected with all three hemoplasmas. There was no association between infection and the different populations. Anemia was associated with Mycoplasma haemofelis and ‘Candidatus Mycoplasma haemominutum’, but not with ‘Candidatus Mycoplasma turicensis’. Male cats and cats with outdoor access were more likely to be infected. Although ‘Candidatus Mycoplasma haemominutum’ is believed to cause minimal or no hematological alterations, the infected cats studied herein were more likely to be anemic.


Três espécies de hemoplasmas são reconhecidas em gatos domésticos: Mycoplasma haemofelis, ‘Candidatus Mycoplasma haemominutum’ e ‘Candidatus Mycoplasma turicensis’. A prevalência e alterações hematológicas associadas à infecção por hemoplasmas foi estudada, em 369 gatos domésticos de três populações distintas (doadores de sangue, hospitais e gatos de abrigo) do Sul do Brasil. Foram realizados hemogramas completos no momento da coleta de sangue e as amostras tiveram seu DNA extraído e testado por PCR convencional para cada espécie de hemoplasmas. Setenta e nove amostras (21,40%) foram positivas para pelo menos uma espécie. O mais prevalente foi ‘Candidatus Mycoplasma haemominutum’ com 50/369 (13,55%) gatos positivos, seguidos por ‘Candidatus Mycoplasma turicensis’ com 10/369 (2,71%) e Mycoplasma haemofelis com 8/369 (2,16%). Coinfecção por Mycoplasma haemofelis e ‘Candidatus Mycoplasma haemominutum’ foi observada em 4/369 (1,08%), enquanto ‘Candidatus Mycoplasma haemominutum’ e ‘Candidatus Mycoplasma turicensis’ coinfectaram 5/369 (1,35%) gatos. ABSTRACT

Systemic lupus erythematosus (SLE) is per se a disease characterized by suppressed immune response and thus susceptibility to various opportunistic infections. We describe the case of a 21-yearold woman who developed a rare zoonosis - hemotrophic mycoplasma infection in the initial stage of SLE, complicated with Nocardia asteroides pneumonia afterwards. Nocardia infection coincided with initiation of glucocorticoids and cyclophosphamide therapy for SLE. After the treatment she recovered completely. To our knowledge the only case of human hemoplasmosis (then referred to as eperythrozoonosis) in medical literature was the one described by a group of Croatian authors 22 years ago. No cases of a hemotrophic mycoplasma infection in a SLE patient have been published up to now.

ABSTRACT

Recent studies have been conducted in Brazil using molecular techniques for the detection of hemotrophic mycoplasmas in several mammals. In domestic cats, Mycoplasma haemofelis, 'Candidatus M. haemominutum', and 'Candidatus M. turicensis' infections have been identified. These species have also been found in free-ranging and captive neotropical felid species. Two canine hemoplasmas, Mycoplasma haemocanis and 'Candidatus Mycoplasma haematoparvum', have been identified in dogs. In commercial swine populations, Mycoplasma suis was found to be highly prevalent, especially in sows. Moreover, novel mycoplasma species have been identified in Brazilian commercial pigs and domestic dogs. A hemoplasma infection in a human patient infected with the human immunodeficiency virus (HIV) was also recently documented. In conclusion, hemoplasma species are common and important infectious agents in Brazil. Further studies should be conducted to better understand their impact on pets, production animals, and wildlife fauna, as well as their role as zoonotic agents, particularly in immunocompromised patients.


Estudos recentes utilizando técnicas moleculares para a detecção de micoplasmas hemotróficos em diferentes mamíferos têm sido conduzidos no Brasil. Em gatos domésticos, infecções por Mycoplasma haemofelis, 'Candidatus M. haemominutum' e 'Candidatus M. turicensis' foram identificadas. Estas espécies também foram encontradas em felídeos neotropicais de vida livre e de cativeiro. Dois hemoplasmas caninos, Mycoplasma haemocanis e 'Candidatus Mycoplasma haematoparvum', foram identificados em cães domésticos. Em populações comerciais de suínos, Mycoplasma suis possui alta prevalência, especialmente em porcas. Além disso, novas espécies de hemoplasmas foram detectadas em suínos comercias e cães. Infecção por um hemoplasma em um paciente humano infectado com o vírus da imunodeficiência humana (HIV) foi recentemente documentada. Em conclusão, espécies de hemoplasmas são comuns e importantes agentes de infecções no Brasil. Estudos futuros devem ser conduzidos para melhor entender seu impacto em cães e gatos, animais de produção e na fauna silvestre, e também para determinar o seu papel como agentes zoonóticos, particularmente em pacientes imunocomprometidos.
